ATP (Adenosine Triphosphate) serves as the fundamental carrier of energy conversion within biological organisms, and its fluctuation directly correlates with the energy metabolism of various organs. As the most crucial energy molecule, ATP plays a pivotal role in various cellular physiological and pathological processes. Alterations in ATP levels can significantly impact the functionality of many cells. Typically, cellular apoptosis, necrosis, or exposure to certain toxic states lead to a decrease in ATP levels, while stimuli such as high glucose can upregulate intracellular ATP levels in some cells. A decrease in ATP levels usually indicates impaired or decreased mitochondrial function, and during cellular apoptosis, the decline in ATP levels often occurs simultaneously with a decrease in mitochondrial membrane potential. The ATP content determination assay kit can be used to detect ATP levels in common solutions, cells, or tissues.

This assay kit is developed based on the catalysis of firefly luciferase, also known as luciferase, which requires ATP to generate fluorescence. When firefly luciferase and luciferin are both present in excess, fluorescence production within a certain concentration range is directly proportional to the ATP concentration. This enables highly sensitive detection of ATP concentration in solutions.

  • Simple sample preparation: no need for perchloric acid or trichloroacetic acid (TCA) extraction, the kit provides a special lysis solution, the sample can be detected for ATP assay after simple lysis.

3. Sample Stability Testing

Conclusion

  1. From Fig. 1: The ATP content of different cells was detected after lysing with MeilunBio and the control product, and the detection value of MeilunBio product was basically better than that of the control product.
  2. From Fig. 2: The stability of MeilunBio product is better than that of the control when the ATP content in the sample is continuously measured for 30 min.
  3. From Table 1: The results of meilunBio and the control product are similar when the same concentration of ATP solution is detected; however, when the same number of cells are detected, the results of MeilunBio product are higher and the data are more accurate.
  4. From Fig. 3 & 4: MeilunBio products have good linearity in the range of 0.1nM-10μM and 0.1nM-100nM ATP concentration when the standard was added.
  5. From Fig. 5: The lysis effect of MeilunBio product was better than that of the control product in WB after lysing HL-60 cells using MeilunBio product and the control product.
