Description
PMA, also known as Phorbol 12-myristate 13-acetate, is an analog of diacylglycerol (DG). It activates protein kinase C (PKC) by binding to its C1 domain, resulting in a cascade reaction similar to the “double messenger system.” In signal pathway research, PMA can be used in combination with PHA/Ionomycin to stimulate cytokine production in cells. It can also be used in conjunction with protein transport inhibitors such as Monensin and Brefeldin A to study the retention of cytokines within cells. PMA can act as a tumor promoter in both in vivo and in vitro tumor research. High concentrations of PMA, with extended treatment time, can downregulate total PKC activity and lead to cell carcinogenesis, while low concentrations can have protective effects. PMA also promotes hematopoietic cell differentiation and can be used to treat human leukemia and neutropenia. With a water solubility 10 times greater than phorbol-12,13-dibutyrate (TLC), PMA is more suitable for cell-based research.
Performance
Stimulate THP-1 cells with PMA to induce differentiation into macrophages; the macrophages then adhere to the wall and exhibit a more regular circular or elliptical shape. (The left figure is the control without PMA, and the right figure is with MeilunBio PMA added)
