LysoTracker Red (LysoTracker Red DND-99) is a paraformaldehyde fixable probe that concentrates into acidic compartments of cells and tissues. LysoTracker Red is an indicator of cell death in a manner similar to other standard assays (Annexin, terminal dUTP nick end labeling, Nile blue sulfate, neutral red, and acridine orange). LysoTracker Red is a marker for late endosomes and lysosomes.
These probes have several important features:
- Selectively label acidic organelles;
- Effective labeling of live cells at nanomolar (nM) concentrations;
- Multiple color probes are available, allowing multi-label experiments based on the situation. The structure of LysoTracker consists of a fluorescent group and an attached weak base, which can freely cross the cell membrane and generally accumulate in spherical organelles, suitable for observing lysosomal internal biosynthesis and related pathological mechanisms. LysoTracker is only partially protonated at neutral pH, so the principle of labeling organelles may be related to its full protonation and retention on the organelle membrane.
