RPMI 1640 medium is named after the Roswell Park Memorial Institute (RPMI), where it was developed. The number 1640 serves as its identification code. It is an improved version of McCoy’s 5A medium, utilizing a bicarbonate buffering system. Initially developed for the suspension or monolayer culture of human leukemia cells, RPMI 1640 medium has been found to be suitable for a variety of mammalian cells, including HeLa, Jurkat, MCF-7, PC-12, PBMC, astrocytes, and cancer cells. It is particularly well-suited for the cultivation of suspension cells and is one of the most widely used culture media available.

The main difference between RPMI 1640 medium and other culture media lies in its inclusion of reduced glutathione and high concentrations of vitamins. RPMI 1640 medium contains biotin, vitamin B12, and folic acid, which are not present in EMEM and DMEM. Additionally, it contains high concentrations of choline chloride and inositol.
