M199 Medium

Product Description

Medium 199 (commonly abbreviated as M199) was first designed by Morgan and colleagues in 1950 for the nutritional study of chicken embryo fibroblasts. It has since found widespread use in culturing various animal cells, including some non-mammalian cells. Its broad species compatibility makes it especially suitable for non-transformed cell cultures. M199 is widely used in virology, vaccine production, and in vitro cultures of primary explants from mouse pancreatic epithelium and rat lens tissue.

Compared to other basal media, M199 contains unique components such as adenine, adenosine, hypoxanthine, thymidine, and other vitamins. M199 does not contain proteins, lipids, or growth factors, so it requires supplementation, typically with serum or serum-free additives. M199 uses a bicarbonate buffering system (2.2 g/L), necessitating a 5-10% CO2 environment to maintain physiological pH.

Production process: This product is prepared by Water-For-Injection (WFI), and the production procedure is strictly in accordance with the cGMP management regulations, and is completed in the clean room for the record of in vitro diagnostic reagents. Performance

1. Promote growth of cells commonly used in virology and vaccine production

3T3-L1, CEF and Vero were inoculated into 12-well plates at 3E+04cells/well, 3E+04cells/well, and 1E+04cells/well, respectively. The cell number and survival rates were detected at 48h of culture under the condition of adding 3% FBS, using two medium, MeilunBio M199 and international brand M199, respectively. At the same time, the uncounted part was changed to the corresponding 1% FBS medium, and the cell number and survival rate were measured again after 48h of further incubation. As shown in the table below, the proliferation and survival rate of 3T3-L1, CEF and Vero cells cultured in MeilunBio M199 medium were higher than those G brand M199 medium.

2. Cell morphology

3T3-L1, CEF and Vero cells were cultured with MeilunBio M199 medium and G brand M199 medium respectively for 72h, and the cell morphology was observed under the microscope.

As shown in the figure below, 3T3-L1 and CEF showed fibroblast-like and Vero showed epithelial-like, and the morphology of the three types of cells cultured in MeilunBio M199 medium and G brand was consistent.
