FireflyGlo Luciferase Reporter Assay System is a glow type quantitative assay kit with high sensitivity and stable luminescent signal, which can meet the high throughput detection of luciferase expression in mammalian cells. FireflyGlo Luciferase Reporter Assay Kit has the following advantages: a new luciferase detection substrate is used, which improves luminescence signal Stability, nearly 2 hours of signal half-life for the experimental design provides greater flexibility; the use of the sample – mixing – detection of the operation method, do not need to rely on the autosampler, and do not need to discard the culture fluid, centrifugation and other steps to simplify the experimental process; the components of the kit has been optimized, compared with the traditional flash-type products, there is no obvious irritating odor. FireflyGlo Luciferase Reporter Gene FireflyGlo luciferase Reporter Gene Kit can be used in a variety of commonly used cell culture solutions: RPMI1640, DMEM, MEM-α, F12, DMEM/F12, etc., with a half-life of about 2 hours (22 ℃), which meets the needs of the majority of high-throughput experiments.

(A) Three-dimensional plot of luciferase concentration – luminescence intensity – time scale curve. Within a certain concentration range, the standard curve statistics show a cross-section, indicating that meilunbio’s FIREFLYGLO luminescent system has good linearity at any moment within 2h. (B) Meilunbio’s FIREFLYGLO Luciferase Reporter Gene Assay Kit is very close in absolute luminescence intensity and stability when compared to the latest version from company P. (C) The single-moment luminescence intensity – Firefly luciferase concentration standard curve has good linearity (R2 = 0.9994).

MeilunBio FIREFLYGLO luminescence system can emit light normally in common culture media, such as DMEM, RPMI1640, DMEM/F12, MEMα, F12, etc. The half-life of the luminescence is around 100-120 min, which is comparable to the official data of the imported brand of P (not shown here, please refer to the official webpage of P Company).

PBS solutions containing a concentration gradient were prepared, and equal amounts of firefly fluorokinase were added to each and the luminescence stability of FIREFLYGLO was verified. The graph shows that Meilunbio® FIREFLYGLO has better phenol red tolerance than the P company brand.

Firefly Luciferase luminescence was detected by in situ tracking in a cell culture medium. As can be seen from the figure, the luminescence stability and absolute light intensity did not differ much from the P company brand; moreover, the light intensity – cell number curve at a single moment had good linearity (R2 = 0.9993).

PBS solutions containing a concentration gradient were prepared, and equal amounts of firefly fluorokinase were added to each and the luminescence stability of FIREFLYGLO was verified. The graph shows that Meilunbio® FIREFLYGLO has better phenol red tolerance than the P company brand. (A) After normalization treatment, it can be seen from the figure that the TNFα concentration gradient maintains a relatively consistent induction relationship at all moments up to 2h; and there is no significant change in this induction relationship as time proceeds. (B) Single moment TNFα concentration gradient induced NF-κB pathway effect of MeilunBio vs. P company brands, and the results were not significantly different.
