Product Description
The Nano-light luciferase Reporter Gene Detection System is a glow-type quantitative test kit with high sensitivity and a stable luminescent signal, which meets the needs of high-throughput testing. Nano-light luciferase is a genetically engineered small molecule enzyme (19.1 kDa) that is a bioluminescent reporter gene of superior performance. It uses a novel substrate that produces high-intensity, glow-type luminescence. The signal intensity is 150 times higher than that of Firefly and Renilla, and the signal stability is superb, with a half-life of up to 2 h. The bioluminescence reaction of Nano-light luciferase is ATP-independent, with a low autoluminescent background and a brighter light signal, while suppressing background luminescence for maximum detection sensitivity.
Nano-light Luciferase Assay System provides a simple, single-addition reagent that generates a glow-type signal in the presence of Nano-light luciferase. The reagent is prepared by mixing Nano-light Luciferase Assay Substrate and Nano-light Luciferase Assay Buffer and contains an integral lysis buffer to use directly on cells expressing Nano-light luciferase or culture medium when the luciferase is secreted.
Performance
Figure 1. NANOLIGHT Luciferase luminescence, stability and luminescence linearity compared to P company brands.
In the in vitro NANOLIGHT Luciferase activity assay of MeilunBio Kit, the absolute luminescence and stability have been very close to the comparison of P company brands, and the NANOLIGHT luminescence linearity is good (R2=0.999)
Figure 2. Validation of luminescence stability and cell density-light intensity standard curve in cell culture system.
In the in vitro NANOLIGHT Luciferase activity assay of MeilunBio Kit, the absolute luminescence and stability have been very close to the comparison of P company brands, and the NANOLIGHT luminescence linearity is good (R2=0.999). In situ detection of luminescence in the cell culture medium showed that the stability and absolute light intensity of the MeilunBio group did not differ much from the P company brand, and, moreover, the cell density-light intensity standard curve was very linear (R2=0.999).
Figure 3. TNFα gradient-induced NF-κB signaling pathway detection effect compared to P company.
The results obtained by MeilunBio Nano-Light Luciferase Reporter Assay Kit maintained good consistency with P company brand.
Figure 4. Validation of NANOLIGHT phenol red tolerance and luminescence in different media.
Phenol red PBS solution containing a concentration gradient was prepared and equal amounts of NANOLIGHT luciferase and luminescence reaction solution were added to each well to verify the luminescence stability of NANOLIGHT. As can be seen from the graph, meilunbio® NANOLIGHT has better phenol red tolerance than the P company brand. In addition, the luminescence level of NANOLIGHT does not fluctuate much in common culture media (normalized by RPMI-1640 in the graph).
