The CFDA SE Cell Proliferation and Tracer Assay Kit is based on a novel fluorescent probe, CFDA SE (Carboxyfluorescein diacetate, succinimidyl ester), which is used for the detection of cell proliferation and cell fluorescence tracing.

The main working principle of the kit is as follows: CFDA SE is a derivative of fluorescein diacetate (FDA), which is a fluorescent dye that can penetrate cell membranes, and does not have fluorescent luminescence itself. When it enters into the living cells through passive transport, it can be catalyzed by esterases in the cytoplasm to generate carboxyfluorescein succinimidyl ester (CFSE), which can emit strong green fluorescence, and the covalently bound fluorescent molecules are rarely shed from the intracellular detachment. During cell division and proliferation, the CFSE-labeled fluorescence can be equally distributed to the two progeny cells, and the fluorescence intensity becomes half of that of the parental cells, so that the CFSE-labeled fluorescence can be detected by flow cytometry.

Therefore, by flow cytometry, depending on the fluorescence intensity, it is possible to detect undivided cells, cells that have divided once (1/2 fluorescence intensity), twice (1/4 fluorescence intensity), three times (1/8 fluorescence intensity), and even more. CFDA SE can detect cells that have divided up to eight times or more. The fluorescence of CFDA SE-labeled non-dividing cells is very stable and homogeneous, and the labeling can be stable for months, making it ideal for cell community analysis.

CFDA SE labeled cells fluoresce green, Ex=492nm, Em=517nm, and can be used in many areas of immunology as a homogeneously stained cell tracer using fluorescence microscopy and flow cytometry. CFDA SE is most commonly used for lymphocyte proliferation assays, but can also be used to detect the proliferation of other cells, such as fibroblasts, natural killer cells, and hematopoietic progenitor cells, among others.

Performance

The fluorescence intensity of cell populations with different numbers of divisions was recorded by flow cytometry on days 0-7 respectively after the labeling of cells with CFDA SE Cell Proliferation and Cell Tracking Kit was completed; and the staining of cells was observed under the fluorescence microscope, and photographs were taken to record the results.

As shown in Figure 1, CFDA SE Cell Proliferation and Cell Tracking Kit labeled the cells well with high fluorescence intensity; as shown in Figure 2, the fluorescence was able to diminish normally with cell division, and the peaks of cells with different division times could be clearly distinguished in the proliferation peaks maps fitted by flow cytometry.
