Product Description

Bio-FireflyGlo Luciferase Reporter Gene Detection System is a glow-type quantitative detection kit with high sensitivity and stable luminescence signal, which can meet the high-throughput detection of luciferase expression in mammalian cells. This product has higher detection sensitivity and stronger luminescent signal than FireflyGlo Luciferase Reporter Assay Kit (MA0519), and can be used to detect the expression of the ADCC Reporter Bioassay or the firefly luciferase reporter gene in cells.

Performance

Figure 1. Comparison of Bio-FIREFLYGLO luminescent system with company P in terms of light intensity and linearity (luciferase level detection).

(A) The absolute light intensity of Bio-FIREFLYGLO Luciferase Reporter Gene Detection Kit surpasses company P. (B) Standard curve of light intensity of Firefly luciferase concentration at a single moment has good linearity (R2=0.9996).

Figure 2. Comparison of Bio-FIREFLYGLO luminescent system with company P in terms of light intensity and linearity (cell level detection).

(A) Cell concentration gradient light intensity stability and (B) Cell concentration-light intensity standard curve example. In a cell culture medium environment, the luminescence of Firefly Luciferase was tracked and detected in situ. Bio-FIREFLYGLO surpasses company P in terms of absolute light intensity; moreover, Standard curve of the light intensity-cell number at a single moment has good linearity (R2=0.9998).

Figure 3. Luminescence of Bio-FIREFLYGLO luminescent system in different culture media.

The MeilunBio Bio-FIREFLYGLO luminescent system can emit light normally in commonly used culture media such as DMEM, RPMI1640, and DMEM/F12, and the results are similar to the control.

Figure 4. Detection effect of 293T cells, inducing NF-κB pathway by TNFα concentration gradient, Meiluncell Bio-FIREFLYGLO compared with company P.

After normalization, the detection results were not significantly different from those of company P.

Figure 5: Detection of luciferase luminescence using PWL110.

Jurkat-luc2p-NFAT-RE-hyg cells were treated with PMA and ionomycin to activate firefly luciferase. PWL110 was used to detect the luminescence after treatment at different drug concentrations, and achieved EC50=3.278ug/ml.

Figure 6: ADCC effect detection of CD20 antibodies using PWL110.

The ratio of effector to target cells is 6:1, EC50=34.76ng/ml. The target cell is Raji, and the effector cell is Jurkat-luc2p-NFAT-RE- FcγRIIIa-high affinity (V158).

Figure 7: AML12 cells were transfected with IL6-promoter-Luc luciferase reporter plasmid, and LPS was added to induce the IL-6 inflammatory pathway.

Compared with company P, Meiluncell Bio-FIREFLYGLO showed no significant difference in results.
